I am working on a project to analyze for residual antifoam in vaccines via Silicon. The antifoam is a polydimethylsiloxane (PDMS) emulsion prepared from a 30% PDMS emulsion. This antifoam is 3% and is further added to the vaccine resulting in a final 0.1% antifoam max. We are trying to analyze down to about 35 ppb Si in solution by ICP-OES and our method is working just fine but we are struggling to get a digestion to work. Currently we have worked with nitric and HF in a microwave digestion but this is not viable for us due to contamination that we cannot remove. So we have moved to a hot block digestion but cannot get recoverable Si. In looking at your notes on this exact problem we see the alcoholic KOH with Xylene but we do not have the sample intro to analyze this matrix. It is not quite clear if there is a way that we can predigest using KOH and then continue with nitric and/or HF to have an aqueous test sample. The notes online seem to suggest a few options but we are having trouble parsing the information and choosing concentrations for reagents. Is this chemistry effective and do we need to use alcoholic KOH or just KOH in water followed by nitric or HF? Can you suggest what concentration of KOH is needed initially to unzip the PDMS if we are working with a potential of up to 50 mg of PDMS in our digestion vessel?
